I have a 2010 C5 Exclusive with the NaviDrive 3D navigation system. The navigation system is not working. Thinking I needed to update the maps etc I bought the latest discs and updated the system but it still not working. I’ve notice that there is a slot for an SD card at the bottom of the unit and the SD card is missing. Do I need this SD card to make the system work and if so, what SD card do I need to get.

No. The SD card is simply for playing media files - it has no other function.
What is not working with the system - you didn't say? Is it not powering on, not booting, no radio, no Navigation - or just a blank screen?
Updating the maps would not do anything if the unit is not working. If anything, reinstalling the firmware may help, but it really does depend on what the issue is.
If you have the screen working and it is just the navigation that is not working - exactly what is not working with the navigation? If you have the other functions and screens working please advise. Resetting the system may be an option if you haven't tried it.

The sat Nav will accept a destination but tell me it’s 500 miles away when maybe only 30 and the map has my location in the middle of the sea. It’s as though the GPS can’t locate me.

All other functions on the system are working such as phone, radio etc.

Ok, that's making more sense.
First things first, your firmware is well out of date. The last firmware is N42C. I can help you with those if you want. Get the firmware sorted first, then you probably also need to turn OFF the 'Synchronise Time with GPS' option.
You may not be aware that the GPS 'N' Week rollover bug will affect certain GPS systems. Read all about that here:

Suffice it to say, that you need to turn this function OFF to prevent miscalculation - it's the only fix to resolve the time issues in addition to what I've mentioned.
See what happens after you have updated the Firmware, as you may have a GPS aerial fault that is preventing the system from locking on to the satellites.
